Hyphal tips from four German isolates of Phytophthora infestans were analyzed for Feulgen DNA content and chromosome number. Nuclear DNA contents were determined following Feulgen staining using scanning integrating microdensitometry. These values were then compared to actual chromosome numbers within hyphal tip nuclei. Chromosome preparations were made by initially treating the tips with colchicine followed by aceto-orcein staining. Chromosome numbers determined during mitotic stages were consistent with mean nuclear DNA contents for each isolate. Scanning integrating microdensitometry has proven to be a reliable method for determining nuclear DNA contents.
