Abstract

The use of the DNA-binding fluorochrome DAPI as a tool in the diagnosis of Lethal Disease of coconut palm in Tanzania is complicated by the occurrence of fluorescence in unstained sections of diseased palms. This autofluorescence is attributed to collapsed phloem elements and phloem and parenchyma cells filled with biogenic amines. It can be distinguished from the DAPI-induced fluorescence with the fluorochrome Rhodamine B. Die Anwendung des Fluorochroms DAPI zur Diagnose der Letalen Erkrankung von Kokospalmen in Tanzania wird kompliziert durch Autofluoreszenzerscheinungen in ungefarbten Schnitten kranker Palmen. Diese Eigenfluoreszenzen sind vermutlich auf kollabierte Siebzellen zuruckzufuhren. Eine Differenzierung der Eigenfluoreszenzen von DAPI-Fluoreszenzen war mit Hilfe des Fluorochroms Rhodamin B moglich.
